A man who was driving the white Ford Kuga, and a woman also travelling in the car, died in the collision on the A500 at Barthomley near Crewe.
Cheshire Police said emergency services were called at about 18:30 GMT on Saturday.
The force added that the next of kin of those who died had been contacted, and appealed for information from people who had been in the area.
Pictures posted on social media showed a fire in the area of the crash as emergency crews responded.
In a statement on Saturday night, police said: "Road closures are in place between the M6 junction 16 and Meremoor Moss Roundabout at Weston. Delays are to be expected and motorists are advised to avoid the area."
